Influencing factors of relative telomere length in workers occupationally exposed to organophosphate

  Objective: The study was designed to assess the change of relative telomere length(RTL) in workers occupationally exposed to organophosphate pesticides(Ops) and determine the effect of telomere binding proteins(TBPs) related genetic polymorphisms and mRNA expression levels.Methods: 180 long-term Ops-exposed workers and 115 healthy controls were recruited.The levels of RTL of DNA in peripheral white blood cells and TBPs including POT1, TRF 1, TRF2, TIN2,RAP1, TPP1 mRNA profiles were analyzed by SYBR Green-based quantitative PCR.Genotyping of POT1, TRF1, TERT was performed with polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) technique.Multiple regression analysis was performed to assess the influencing factors of RTL.Results:The workers 1.50(1.11, 2.57) had significantly higher RTL than controls 0.95(0.76,1.32).There were no significant correlations between POT1 rs10250202、 POT1 rs1034794、 TERT rs2736098 and TRF1 rs3863242 and RTL for Ops-exposed workers or controls.The workers had significantly lower POT1, RAP1, TIN2, TPP1, TRF1,TRF2 mRNA expression levels than controls.POT1mRNA, RAP1 mRNA and TIN2 mRNA levels negatively correlated with RTL(P<0.05), and TPP1 mRNA, TRF1 mRNA levels positively correlated with RTL(P<0.05).Multiariable analysis indicated Ops exposure(b=-1.110, P<0.001), gender(b=0.296, P =0.007), TRF1 rs3863242(b=0.215, P =0.011), TIN2 mRNA(b=-0.738, P =0.013), POT1 mRNA(b=1.367, P <0.001), TRF1 mRNA(b=0.233, P <0.001), TPP1 mRNA(b=0.132, P <0.001) could influence RTL.Conclusions: Occupational Ops exposure may cause workers telomere length extension.Ops exposure, and gender,TRF1 rs3863242, TIN2 mRNA, POT1 mRNA, TRF1 mRNA, TPP1 mRNA are potential susceptibility biomarkers of chromosomal damage in Ops-exposed workers.
