The graded glucose infusion (GGI) procedure has been used to evaluate β cell function independently of insulin sensitivity in humans and in the present study was used to quantify β-cell function in cynomolgus monkeys (Macacafascicularis) ranging from normal to overtly diabetic.Quantification of insulin secretion rate (ISR) using the GGI provides insight into the trajectories ofβ-cell dysfunction in the natural development of diabetes and is an important method to directly quantify the effects of therapeutic agents on β-cell function.ISR was determined by deconvolution of serum C-peptide concentrations measured during a 5 stage GGI (glucose infusion rates of 2, 4, 8, 16, and 32 mg/kg/min for 40 mins each) in 6 normal monkeys (6.0 ± 0.2 yrs, 7.6 ± 0.7 kg, fasting glucose: 60 ± 4 mg/dL), 14 prediabetic monkeys (15.0 ± 0.8 yrs, 9.0 ± 0.9 kg, fasting glucose: 80 ± 4 mg/dL), and 4 diabetic monkevs not treated with insulin (16 ± 1.2 yrs, 9 ± 1.8 kg, fasting glucose: 169 ± 22 mg/dL).
