Cloning, expression and characterization of cysteine protease gene from Euphorbia helioscopia

  The full-length cDNA encoding a putative papain-like cysteine protease was isolated from Euphorbia helioscopia (EhCP,Genebank accession number: KP995938) by the rapid amplification of cDNA ends (RACE) method.The cDNA contained an open reading frame of 1083 bp corresponding to a polypeptide of 361 amino acids with a predicted moleculer weight of 40.48 kDa.Homology of EhCP protein with cysteine protease of Ricinus communis,Jatropha curcas and Populus trichocarpa was 75%,74% and 73% respectively.Phylogenetic tree analysis showed that the EhCP had the highest identity and the closest genetic relationship with Jatropha curcas,Ricinus communis,Populus euphratica,and Populus trichocarpa.Transcription pattern analysis revealed that EhCP had high transcription in laticifer,and low transcription in bark and leaf.Results obtained by real-time RT-PCR analysis indicated that the EhCP expression in root was lowest,and that in leaf and stem were similar.Cloning of the EhCP gene will enable us to further understand the possible function in the laticifer development of E.helioscopia.
