One of the central questions facing breast tumor biology is how cancer cells activate telomerase, which in the great majority of cases is required for unlimited cell growth.Two conflicting theories have emerged so far.One states that malignant cells arise from telomerase-negative normal somatic cells, and, in order to immortalize, must induce hTERT gene encoding the catalytic rate-limiting subunit of telomerase.According to the other theory, malignant cells arise from telomerase-positive stem cells.Recently published data as well as our preliminary experiments, however, indicate that the picture may be significantly more complex.In vivo, many normal breast epithelial cells express significant levels of hTERT, while hTERT levels in DCIS and invasive carcinomas are heterogeneous and in many cases lower than in normal tissues.These data suggest that telomerase that is active in normal breast epithelial cells is inactivated during early cancer progression and is reactivated during later stages of the disease.
