The view that tumor metastasis and recurrence are caused by self-renewal and propagation of cancer stem cells is supported by a series of recent studies.Originally cloned as a dominant inhibitor of the hyperactive EGFR mutant Elipse the cell fate determination factor, DACH1 is a key member of the Retinal Determination Gene Network.We have previously reported that loss of DACH1 expression, examined in >2,000 human breast cancers, correlated with poor prognosis in breast cancer.DACH 1 inhibits breast cancer cell DNA synthesis, colony formation, and tumor growth in vivo.Rather than simply blocking cellular proliferation, DACH 1 reverses the transformed phenotype of transformed breast epithelial cells in 3-dimensional culture.Thus, DACH 1 represents a new type of breast tumor inhibitor, although the mechanism remains unclear.
