Eradication of Brain Tumor Stem Cells through Oncolyticadenovirus-mediated Autophagic Cell Death

　　Malignant astrocytic gliomas such as glioblastoma are the most common and lethal intracranial tumors.Despite implementation of intensive therapeutic strategies and supportive care, the median survival of patients with GBM has remained at 12 months for the past decade.Efficacious and targeted therapies are urgently needed.In the fullness of time, oncolytic adenoviruses may prove to be crucial for the therapy of malignant gliomas.Delta-24-RGD, an E 1A mutant oncolytic adenovirus developed in our laboratory that is being tested in the clinical setting, has shown prominent efficacy in glioma cell lines and intracranial glioma models as well as in other types of tumor.Recently, we reported that this virus was an efficient agent against brain tumor stem cells (BTSCs) with an abnormal p 16INK4/Rb pathway that are resistant to radiation and chemotherapy, and are therefore proposed to account for tumor recurrence.
