  Most of the amino acids have an asymmetric carbon at the alpha position.Although the L-amino acids are abundantly present in higher animals, the minor component, D-amino acids are currently gradually recognized as the candidates of physiologically active substances and biomarkers.In mammals including human beings, several D-amino acids, such as D-Ser, D-Asp, D-Ala, D-Leu and D-Pro, were found and a sensitive and selective analytical method for the simultaneous determination of all proteinogenic amino acid enantiomers is highly required.To accomplish this aim, we have established an enantioselective micro-2D-HPLC system for all proteinogenic amino acids.Amino acids were derivatized with a highly sensitive fluorescent reagent, 4-fluoro-7-nitro-2,1,3-benzoxadiazole (NBD-F), and the derivatives were determined using a micro-2D-HPLC system combining a microbore-reversed-phase column (1st dimension) and a set of narrowbore-enantioselective columns (2nd dimension).
