Current methods for cell-based screening often use immortalized cell lines and genetically engineered expression of target genes and/or reporter constructs.While these approaches ease the burden of increasing throughput through simplified assay protocols,they often feature false positives and false negative which can dramatically reduce the success rate of screening campaigns.Described here is a novel approach to cell based assays that is fully automatable and integrates with existing liquid handling robotics.Heterogeneous assays such as immunocytochemistry can be run in highly miniaturized formats,allowing primary cells to be used and avoids the risk of artifacts associated with genetic engineering and reporter genes.Another hallmark of advanced cell models,3-dimensional extracellular matrix,can be readily incorporated into cellular assays using this approach.Also,because of the inherent customization capabilities of this platform,functional assays such as those based on cell motility and even gradients become easily automatable.Assays including angiogenesis,differentiation,epithelial to mesenchymal transition,leukocyte chemotaxis and tumor cell invasion will be highlighted.
