目的探讨膜相关蛋白Numb在上皮性卵巢癌中的表达及其与卵巢癌干细胞标志物CD117、CD133、乙醛脱氢酶1(ALDH1)的关系。方法选择四川大学华西第二医院妇科住院的136例卵巢肿瘤患者及无卵巢肿瘤患者22例为研究对象,根据摘除卵巢的病理组织学检查结果,将其分为上皮性卵巢癌组(n=92)、交界性卵巢肿瘤组(n=23)、良性卵巢肿瘤组(n=21)及正常卵巢组(n=22)。采用免疫组化SP法检测患者卵巢组织中Numb、CD117、CD133及ALDH1蛋白的表达水平,统计分析几种蛋白的表达差异及相关性。结果 1Numb蛋白在上皮性卵巢癌组阳性表达率高于良性卵巢肿瘤组及正常卵巢组,交界性卵巢肿瘤组Numb蛋白阳性表达率高于正常卵巢组,且差异均有统计学意义(P<0.05)。2Numb蛋白阳性表达率在上皮性卵巢癌患者中FIGO分期为Ⅰ~Ⅱ期患者低于Ⅲ~Ⅳ期患者,无淋巴结转移患者低于有淋巴结转移患者,且阳性表达率差异均有统计学意义(P<0.05);而不同年龄、组织病理学类型、病理分级及残留灶大小之间比较,差异则无统计学意义(P>0.05)。3上皮性卵巢癌患者卵巢组织中Numb蛋白与CD117及CD133阳性表达无相关性(r=0.116,P=0.261;r=0.083,P=0.425);而Numb蛋白阳性表达与ALDH1表达呈正相关关系(r=0.296,P=0.003)。结论 Numb蛋白的表达与上皮性卵巢癌的发生、侵袭和转移有关,与卵巢癌干细胞标志物ALDH1表达呈正相关,有可能影响患者的预后。
Objective To investigate the expression of membrane-associated protein Numb in epithelial ovarian cancer and its relationship with CD117, CD133 and ALDH1. Methods Totally 136 patients with ovarian cancer and 22 patients without ovarian cancer who were hospitalized in Gynecology Department of West China Second Hospital of Sichuan University were enrolled in this study. According to the histopathological findings of ovariectomized patients, they were divided into epithelial ovarian cancer group (n = 92) ), Borderline ovarian tumor (n = 23), benign ovarian tumor (n = 21) and normal ovary (n = 22). The expression of Numb, CD117, CD133 and ALDH1 protein in ovarian tissue was detected by immunohistochemical SP method. The differences and correlations of several proteins were statistically analyzed. Results The positive expression rate of Numb protein in epithelial ovarian cancer group was higher than that in benign ovarian tumor group and normal ovarian group, and Numb protein in borderline ovarian tumor group was higher than that in normal ovarian group (P <0.05) ). The positive rate of 2Numb protein in patients with epithelial ovarian cancer with FIGO stage Ⅰ ~ Ⅱ was lower than those in patients with stage Ⅲ ~ Ⅳ, the patients without lymph node metastasis were lower than the patients with lymph node metastasis, and the positive expression rates were statistically significant ( P <0.05). However, there was no significant difference between different age, histopathological types, pathological grading and residual tumor size (P> 0.05). There was no correlation between the expression of Numb protein and the positive expression of CD117 and CD133 in ovarian epithelial ovarian cancer (r = 0.116, P = 0.261; r = 0.083, P = 0.425), while the positive expression of Numb protein was positively correlated with the expression of ALDH1 r = 0.296, P = 0.003). Conclusion The expression of Numb protein is related to the occurrence, invasion and metastasis of epithelial ovarian cancer and positively correlated with ALDH1 expression of ovarian cancer stem cell marker, which may affect the prognosis of patients.
