目的:研究鼻脑通鼻用喷雾剂对实验性偏头痛小鼠内皮素-1(ET-1)、降钙素基因相关肽(CGRP)含量的影响。方法:75只ICR小鼠,雌雄各半,随机分为5组,即对照组、模型组、鼻脑通鼻用喷雾剂低、中、高剂量组,每组15只。采用硝酸甘油皮下注射制备小鼠实验性偏头痛模型,造模后20 min开始记录小鼠前肢搔头的次数,记录10 min;造模4 h后腹主动脉取血,测定血浆中ET-1、CGRP指标。结果:与对照组相比,模型组动物搔头次数显著增多(P<0.001);提前给予鼻脑通鼻用喷雾剂3个剂量后,造模小鼠的搔头次数均比模型组显著减少(P<0.05,P<0.01,P<0.001)。与对照组相比,模型组动物血浆ET-1含量显著升高(P<0.001)、CGRP含量升高(P<0.01);而与模型组相比,提前给予鼻脑通鼻用喷雾剂3个剂量后,造模的动物血浆中、高剂量组ET-1含量显著降低(P<0.001,P<0.001),低、中、高剂量组CGRP含量均降低(P<0.05,P<0.01,P<0.01)。结论:鼻脑通鼻用喷雾剂能减少实验性偏头痛动物的搔头次数,其治疗偏头痛的机制可能与降低动物血浆ET-1、CGRP含量有关。
Objective: To study the effects of nasal and cerebral nasal spray on endothelin-1 (ET-1) and calcitonin gene-related peptide (CGRP) in experimental migraine mice. Methods: Seventy-five ICR mice were randomly divided into 5 groups: control group, model group, low, medium and high dose nasal and nasal spray group, 15 rats in each group. The model of experimental migraine was prepared by subcutaneous injection of nitroglycerin. The number of scratching head in mice was recorded at 20 min after model establishment and recorded for 10 min. Blood samples were obtained from the abdominal aorta 4 h after model establishment, and plasma levels of ET-1 , CGRP indicator. Results: Compared with the control group, the number of scratching of animals in the model group increased significantly (P <0.001). After three doses of nasal and nasal sprays were given in advance, the frequency of scratching of the model mice decreased significantly (P <0.05, P <0.01, P <0.001). Compared with the control group, the plasma ET-1 level in the model group was significantly increased (P <0.001) and the content of CGRP increased (P <0.01). Compared with the model group, nasal brain nasal spray 3 (P <0.001, P <0.001). The levels of CGRP in low, medium and high dose groups were significantly decreased (P <0.05, P <0.01, P < P <0.01). Conclusion: Nasal and nasal spray can reduce the frequency of head scratching in experimental migraine animal and the mechanism of migraine treatment may be related to the decrease of plasma ET-1 and CGRP in animals.
