目的 :研究CBN注射剂中葛根素在大鼠体内排泄的特点。方法 :取大鼠尾静脉注射 10 0mg/kg给药后不同时间分别收集尿和粪便。乌拉坦麻醉大鼠做胆总管插管 ,于股静脉给药后不同时间采集胆汁。以HPLC外标法测定样品中葛根素含量 ,计算不同时间自尿粪胆汁排出量及占给药量百分比。结果 :静脉给药后 12h药物自尿排出给药量的 34 6 % ,其中 85 6 %在前 2h从尿中排出 ,12h后尿中几无法测出。粪便中葛根素含量很少 ,胆汁中葛根素在药后 8h时累积排泄量为 12 7μg/ml ,占总排泄量的2 7% ,其中 70 4%于前 2h内排出。结论 :静脉注射CBN后其葛根素原型主要以尿中排出 ,而排出时间集中在药后的前 2h。
Objective: To study the characteristics of excretion of puerarin in rats in CBN injection. Methods : Urine and feces were collected at different time after administration of tail vein injection of 100 mg/kg. Ulantan anesthetized rats underwent common bile duct cannulation, and bile was collected at different times after femoral vein administration. The puerarin content in the sample was determined by HPLC external standard method, and the urinary excretion of bile was calculated at different times and the percentage of the dose was calculated. RESULTS : The urinary excretion of drug was 346 % after intravenous administration for 12 h. 85 % of the drug was excreted in the urine during the first 2 h and could not be detected in the urine after 12 h. The content of puerarin in feces was very small, and the cumulative excretion of puerarin in bile was 8 7 μg/ml 8 h after the drug, which accounted for 27% of the total excretion, of which 70.4% was excreted within the first 2 h. Conclusion: The puerarin prototype was mainly excreted in urine after intravenous injection of CBN, and the discharge time was concentrated in the first 2 h after drug administration.
