在室温下,将虎杖(Poygonum cuspidatum)的干燥地下根以95%乙醇提取,提取液经真空蒸去乙醇后的残渣,置于二氯甲烷-水(1:1)中分配层析。二氯甲烷可溶性组分经己烷-90%水甲醇(1:1)进一步分配层析,得到的甲醇组分在蛋白质酪氨酸激酶(PTK)分析试验中呈现强的抑制活性(IC<80μg/ml)。将活性的95%甲醇组分上硅胶闪蒸柱,用氯仿/甲醇(5%~50%)洗脱,得7个组分(A~G)。活性组分C(IC_(50)<80μg/ml)经浓缩后重结晶
The dried roots of Poygonum cuspidatum were extracted with 95% ethanol at room temperature, and the residue after vacuum evaporation of the extract was placed in dichloromethane-water (1:1) and partitioned and chromatographed. Dichloromethane soluble fractions were further fractionated with hexane-90% aqueous methanol (1:1) and the resulting methanol fraction exhibited strong inhibitory activity in protein tyrosine kinase (PTK) assays (IC<80 μg /ml). The active 95% methanol component was flash chromatographed on silica gel eluting with chloroform/methanol (5%-50%) to give 7 components (A-G).
