目的对江苏省2010年发热伴血小板减少综合征布尼亚病毒(SFTSV)进行核酸检测并分析其全基因组序列。方法用荧光定量PCR的方法对江苏省2010年SFTSV感染病例进行核酸检测,将所有阳性标本进行病毒分离,取8株代表性毒株测定其全基因组序列,序列结果用DNAStar和MEGA4软件进行分析。结果 33例发热伴血小板减少综合征疑似病例中,20例SFTSV核酸检测阳性。1例狗标本SFTSV检测也为阳性。8株SFTSV毒株全基因组序列与国内其他地区流行株的序列高度同源,其中M片段核苷酸序列同源性介于95.8%~98.7%之间,氨基酸序列同源性介于98.7%~99.7%之间;而与汉坦病毒属和白蛉热病毒属的相关序列相比差异较大,M段核苷酸序列同源性最高仅为27.1%,氨基酸序列同源性最高仅为7.3%。与一代病例密切接触后发病的二代病例序列与一代病例序列完全一致;狗携带的SFTSV序列与病人序列高度同源。结论 SFTSV江苏株与国内参考株序列无明显差异,与布尼亚病毒科的其他属序列差异巨大;SFTSV存在人传人的可能性;狗可能是SFTSV的传播宿主之一。
Objective To detect the nucleic acid of fever virus with thrombocytopenic syndrome Bunyavirus (SFTSV) in 2010 and analyze the whole genome sequence. Methods Fluorescent quantitative PCR was used to detect the nucleic acid of SFTSV infection in Jiangsu Province in 2010. All the positive samples were isolated from the virus. Eight representative strains were used to determine the whole genome sequence. The sequence was analyzed by DNAStar and MEGA4 software. Results Of 33 cases of fever with thrombocytopenia syndrome suspected cases, 20 SFTSV nucleic acid test positive. SFTSV test was also positive in 1 case of dog. The complete genome sequences of 8 strains of SFTSV were highly homologous to the sequences of other epidemic strains in other regions of China. The nucleotide sequence homology of M fragments was between 95.8% ~ 98.7%, and the amino acid sequence homology was 98.7% 99.7%, respectively. However, they were significantly different from the related sequences of Hantavirus and Brachyspira. The homology of M segment nucleotide sequence was only 27.1%, and the highest homology of amino acid sequence was only 7.3 %. The sequence of the second-generation case that came into close contact with a generation of cases was exactly the same as the one-generation case sequence. The SFTSV sequence carried by the dog was highly homologous to the patient sequence. Conclusion There is no significant difference between SFTSV Jiangsu strains and domestic reference strains, which is quite different from other genera of Bunyaviridae. SFTSV has the possibility of human transmission. Dogs may be one of the transmission hosts of SFTSV.
