重组可溶性TRAIL诱导肿瘤细胞凋亡

在大肠杆菌系统中表达并纯化了3种不同长度的可溶性TRAIL, 分别为sTRAIL(74~281), sTRAIL (95~281)及sTRAIL(101~281). 细胞凋亡实验表明氨基酸101位前的序列对TRAIL诱导肿瘤细胞凋亡的生物学功能具有抑制作用. sTRAIL(101~281)能在6 h内诱导不同类型的肿瘤细胞凋亡, 但对正常细胞无毒性. 不同类型的肿瘤细胞对TRAIL的敏感程度不同, 一些敏感肿瘤细胞系中部分细胞对TRAIL具有抗性. 根据实验结果提出抗性细胞中可能表达一种或多种半衰期短的、能抑制肿瘤细胞凋亡的蛋白的假说, 并为探索将TRAIL开发成肿瘤治疗药物的可能性奠定了基础. Three different lengths of soluble TRAIL were expressed and purified in Escherichia coli system, which were sTRAIL (74-281), sTRAIL (95-281) and sTRAIL (101-281), respectively. Sequence can inhibit the biological function of TRAIL-induced tumor cell apoptosis.TRAIL (101 ~ 281) can induce different types of tumor cell apoptosis within 6 h, but it is not toxic to normal cells.Different types of tumor cells inhibit TRAIL , Some cells in some sensitive tumor cell lines are resistant to TRAIL.According to the experimental results, it is proposed that the resistant cells may express one or more proteins with short half-life and can inhibit the apoptosis of tumor cells It laid the foundation for exploring the possibility of developing TRAIL into a cancer therapeutic drug.
