目的:观察和比较再生障碍性贫血(AA)、骨髓增生异常综合征(MDS)和正常人来源的骨髓间充质干细胞(MSCs)的形态和生长特征的差异。方法:从AA患者、MDS患者及正常人骨髓中分离MSCs,进行体外培养,观察细胞形态特征,绘制生长曲线描述细胞生长情况。结果:研究发现AA患者的骨髓MSCs的形态与正常对照组无异,均为梭形成纤维细胞样,但AA患者的骨髓MSCs生长缓慢,集落形成时间及细胞融合达90%所需时间均长于正常对照组,P3代细胞增殖能力低于正常对照组。MDS患者的骨髓MSCs的形态、集落形成时间及细胞融合达90%所需的时间与正常组无明显差别,P3代细胞增殖能力与正常正常对照组无异。结论:AA和MDS患者的骨髓MSCs的形态与正常对照组无异,但AA患者骨髓MSCs的生长速度和增殖能力明显低于正常对照组,而MDS患者骨髓MSCs的细胞形态、生长速度和细胞增殖能力与正常对照组相比无明显差别。
OBJECTIVE: To observe and compare the morphological and growth characteristics of aplastic anemia (AA), myelodysplastic syndrome (MDS) and normal human bone marrow mesenchymal stem cells (MSCs). Methods: MSCs were isolated from the bone marrow of AA patients, MDS patients and normal individuals. The MSCs were cultured in vitro. The morphological characteristics of the cells were observed. The growth curve was drawn to describe the cell growth. Results: The morphology of bone marrow MSCs in patients with AA was similar to that of the normal control group, all of them were spindle-shaped fibroblast-like cells. However, the bone marrow MSCs grew slowly in AA patients and the time required for colony formation and 90% cell fusion were longer than normal The control group, P3 generation of cell proliferation was lower than the normal control group. The morphology, colony forming time and the time required for 90% cell fusion of bone marrow MSCs in MDS patients were not significantly different from those in normal group. The proliferation ability of P3 cells was similar to that of normal control group. CONCLUSIONS: The morphology of bone marrow MSCs in patients with AA and MDS is similar to those in normal controls. However, the growth and proliferation of MSCs in AA patients are significantly lower than those in normal controls. However, the morphology, growth rate and proliferation of MSCs in MDS patients There is no significant difference between the ability and the normal control group.
