[目的]探讨整合素α5和含半胱氨酸的天冬氨酸蛋白水解酶3(caspase 3)在肝细胞癌(HCC)中的表达及其临床意义.[方法]收集2015年1月至12月40例在本院实施手术切除并经病理确诊为HCC病例的癌组织石蜡标本以及癌旁组织标本,备行免疫组织化学检查整合素α5、caspase 3于HCC中的表达情况;收集2016年1月至12月期间选择实施手术切除的HCC患者30例的新鲜HCC组织和对应的癌旁组织,备行荧光定量RT-PCR和蛋白质印迹法检测整合素α5、easpase3的mRNA表达.[结果]肝癌组织中整合素α5表达率显著高于,caspase 3的阳性表达率低于癌旁组织(P<0.05);40例HCC患者的整合素α5蛋白质表达与TNM分期和肿瘤转移密切相关(P<0.05),caspase 3的蛋白质表达与HBsAg、TNM分期、肿瘤转移密切相关(P<0.05);.肝癌组织中整合素α5和caspase 3的表达具有负相关性(P<0.05);肝癌组织中的整合素α5 mRNA相对表达量明显高于癌旁组织,caspase 3 mRNA低于癌旁组织(P<0.05).[结论]整合素α5与caspase 3呈负相关表达,其可能在肝癌细胞的凋亡期间进行参与,但它们对于肝癌细胞失巢凋亡的作用机制需要进一步的深入研究.“,”【Objective】To observe and analyze the expression and clinical significance of integrin a 5 and as-partate protease 3 containing cysteine (caspase 3) in patients with hepatocellular carcinoma(HCC). 【Methods】Paraffin-embedded specimens of 40 HCC cancer and paracancerous tissues from January to December 2015 underwent surgical resection in our hospital HCC were collected. Immunohistochemical examination of integrin a 5 and caspase 3 expression was performed. Also, fresh HCC tissues and corresponding paracancerous tissues were collected from 30 HCC patients who underwent surgical resection between January to December, 2016. Fluorescence quantitative RT-PCR and Western blotting were used to detect integrin a 5 and caspase 3 expression. 【Results】The expression rate of integrin a 5 in HCC was significantly higher than that in adjacent tissues, while the expression of caspase 3 in HCC was lower than that in paracancerous tissue ( P <0.05). The expression of integrin a 5 protein in 40 patients with HCC was closely related to TNM stage and tumor metastasis (P <0.05), while caspase 3 protein expression was related to HBsAg, TNM stage and tumor metastasis ( P <0.05). There was a negative correlation between integrin a 5 and caspase 3 expression in HCC ( P <0.05). The relative expression of integrin a 5 mRNA in HCC was significantly higher than that in adjacent tissues. The caspase 3 mRNA in the liver cancerous tissue was significantly lower than that in the paracancerous tissue (P<0.05). 【Conclusion】The negative correlation between integrin a 5 and caspase 3 may be involved in the apoptosis of hepatoma cells. However, the mechanism of integrin a 5 0n apoptosis of hepatoma cells requires further study.
