目的从基因转录水平确定人视网膜色素上皮(RPE)细胞是否具有合成纤溶酶原激活物抑制1(PAI1)蛋白酶的生物学特性。方法取供体人眼RPE细胞进行体外培养;当细胞生长融合后抽取RNA,应用Nothernblot分析技术,经特异的探针对人RPE细胞的总RNA进行检测。结果在体外培养的人RPE细胞基因转录产物中存在有PAI1mRNA的表达,其长度分别为3.0kb和2.3kb。结论人RPE细胞能够合成PAI1蛋白酶;它可能在一些致盲性眼病的病理发生过程中起到重要的作用。
Objective To determine whether the human retinal pigment epithelium (RPE) cells have the biological characteristics of the synthetic plasminogen activator inhibitor-1 (PAI-1) protease from the gene transcription level. Methods Donor human eye RPE cells were cultured in vitro. After the cells grew and fused, RNA was extracted and the total RNA of human RPE cells was detected by specific probe using Nothernblot assay. Results The expression of PAI-1 mRNA existed in human RPE cells cultured in vitro with length of 3.0kb and 2.3kb, respectively. Conclusions Human RPE cells can synthesize PAI-1 protease; it may play an important role in the pathogenesis of some blinding ophthalmias.
