目的检测和分选人肺腺癌细胞系SPCA1中的边缘细胞,并对其表面标志进行初步分析。方法用流式细胞仪检测和分选SPCA1中的边缘细胞和主群细胞,并用细胞免疫化学和激光共聚焦检测ABCG2和CD133在SPCA1中的表达。结果边缘细胞亚群占SPCA1细胞的0.7%,维拉帕米阻断后减少至0.0%。细胞免疫化学结果显示ABCG2和CD133在SPCA1的边缘细胞中高表达,而在未分选的SPCA1细胞和主群细胞中则低表达。结论人肺腺癌细胞系SPCA1中存在边缘细胞,提示肺癌干细胞的存在。
Objective To detect and sort the marginal cells in human lung adenocarcinoma cell line SPCA1 and to analyze the surface markers. Methods Flow cytometry was used to detect and sort the marginal cells and main population cells in SPCA1. The expression of ABCG2 and CD133 in SPCA1 was detected by immunocytochemistry and confocal laser scanning microscope. Results The marginal cell subpopulation accounted for 0.7% of SPCA1 cells and decreased to 0.0% after verapamil was blocked. Immunocytochemistry results showed that ABCG2 and CD133 were highly expressed in the marginal cells of SPCA1, but lower in unsorted SPCA1 cells and in the main population. Conclusion The existence of peripheral cells in human lung adenocarcinoma cell line SPCA1 suggests the existence of lung cancer stem cells.
