本文用指纹法定量分析测定22种氨基酸,为前文“定性分离鉴定”的继续。定位显色剂采用镉—茚三酮酸化丙酮溶液,显色的最适温度及时间分别为50°—55℃及30分钟;再用甲醇洗脱30分钟,然后在波长为505nm处进行比色定量。本法测定氨基酸的灵敏度为10~(-6)—10~(-7)g,回收实验的误差在±10%以内。对8例正常人血清中16种游离氢基酸含量进行了定量分析测定,所得结果的范围与用离子交换柱或双向纸层析法相一致。本法可用于人体体液中游离氨基酸含量的微量定量分析,亦可用来测定蛋白质水解物的氨基酸含量。
