采用改进的细胞移动测量方法,观察巨噬细胞分泌因子对动脉平滑肌细胞移动的直接刺激作用。传代接种的培养细胞以平血中琼脂糖层中心孔为界贴壁形成圆形集落,经照射后分组培养,定期测量细胞集落向外扩展的面积。结果表明,在巨噬细胞条件培养液中,平滑肌细胞每天向外移动约10.6mm~2,在无血小板及全血血清培养液中,分别为5.7及8.6mm~2。实验结果提示巨噬细胞的分泌产物具有直接促进动脉平滑肌细胞移动的化学促动作用。
The improved cell migration measurement method was used to observe the direct stimulating effect of macrophage secretion factor on arterial smooth muscle cell migration. The passaged inoculated cultured cells formed circular colonies adhered to the center hole of the agarose layer in the flat blood and were cultured in groups after irradiation to regularly measure the outward expansion of the cell colonies. The results showed that in macrophage conditioned medium, smooth muscle cells migrated outward by about 10.6mm ~ 2 per day, and in platelet - free and whole blood serum culture mediums, 5.7 and 8.6mm ~ 2, respectively. The experimental results suggest that the secretory products of macrophages have a direct chemical stimulating effect on arterial smooth muscle cell migration.
