目的探究肾母细胞瘤组织中色素上皮衍生因子表达及在肾母细胞瘤血管新生方面的作用,分析其表达水平与临床病理的联系。方法采用免疫组织化学方法检测40例肾母细胞瘤和对照组18例癌旁2 cm肾组织中色素上皮衍生因子和CD34分子的表达,计算肾母细胞瘤组织中的微血管密度并分析其与PEDF表达水平的相关性,分析PEDF的表达水平与临床病理参数的联系。结果肾母细胞瘤组织中PEDF表达较癌旁对照组显著下调,差异有统计学意义(P<0.05)。肾母细胞瘤中PEDF的表达水平与微血管密度成负相关(r=-0.872,P<0.05)。肾母细胞瘤组PEDF蛋白表达水平与患儿年龄、组织分级、临床分级及有无淋巴结转移等多种临床病理因素密切相关(P<0.05)。结论肾母细胞瘤中PEDF蛋白表达与癌旁对照组织相比明显下调,与肾母细胞瘤的发病密切相关。
Objective To investigate the expression of pigment epithelium-derived factor (EGF) in nephroblastoma tissue and its role in angiogenesis of nephroblastoma and analyze its relationship with clinicopathology. Methods Immunohistochemistry was used to detect the expression of pigment epithelium-derived factor and CD34 in 40 cases of nephroblastoma and 18 cases of paracarcinoma adjacent to the control group. The microvessel density in nephroblastoma was calculated and analyzed with PEDF The correlation between PEDF expression and clinical pathological parameters was analyzed. Results The expression of PEDF in nephroblastoma was significantly lower than that in the adjacent non-cancerous tissues (P <0.05). The expression of PEDF in nephroblastoma was negatively correlated with the microvessel density (r = -0.872, P <0.05). The expression of PEDF protein in Wilms tumor was closely related to various clinicopathological parameters such as age, histological grade, clinical grade and lymph node metastasis (P <0.05). Conclusion The expression of PEDF protein in Wilms ’tumor is significantly down-regulated compared with that in adjacent non-cancerous tissues, which is closely related to the pathogenesis of Wilms’ tumor.
