目的探讨滋阴活血方药(nourishing yin and promoting blood-flow recipe,NYPBR)对糖尿病大鼠牙周组织的保护作用。方法 2007年6-10月于河北医科大学选用6月龄雄性Wistar大鼠28只,随机分为3组:空白对照组(8只)、糖尿病组(10只)和中药治疗组(10只)。除空白对照组外,其余各组大鼠腹腔内注射链脲佐菌素(STZ)溶液,建立糖尿病大鼠模型。模型建立后,中药治疗组灌胃给予NYPBR12周,结束给药后第1天处死大鼠,取上颌骨标本苏木素-伊红(HE)染色,光镜观察牙周组织病理变化。结果糖尿病组大鼠牙龈上皮和固有层中度炎细胞浸润,毛细血管数目增多、扩张充血,纤维排列紊乱、断裂,骨吸收陷窝多,陷窝内见破骨细胞,牙槽嵴顶无吸收;而中药治疗组牙周组织结构与空白对照组相似。结论 NYPBR能显著减轻糖尿病造成的牙周组织损伤。
Objective To investigate the protective effects of nourishing yin and promoting blood-flow recipe (NYPBR) on periodontal tissues in diabetic rats. Methods Sixty-eight male Wistar rats aged 6 months were randomly divided into three groups: control group (n = 8), diabetes mellitus group (n = 10) and TCM treatment group (n = 10) . Except the blank control group, the other rats in each group were injected streptozotocin (STZ) solution intraperitoneally to establish a diabetic rat model. After the model was established, the Chinese medicine treatment group was given gavage NYPBR for 12 weeks, and the rats were killed on the first day after the end of administration. The maxillary specimens were stained with hematoxylin and eosin (HE) and the pathological changes of the periodontal tissues were observed with light microscope. Results In the diabetic group, there was moderate infiltration of inflammatory cells in the gingival epithelium and lamina propria, with an increase in the number of capillaries, dilatation and congestion, disordered fibers, fractures, more lacunae in the lacunae, osteoclasts in the lacunae, and no absorption at the alveolar crest ; While the traditional Chinese medicine treatment group periodontal tissue structure similar to the blank control group. Conclusion NYPBR can significantly reduce periodontal tissue damage caused by diabetes.
