目的:证实脂多糖致新生鼠脑白质软化模型的可靠性。方法:16只健康孕鼠随机分为实验组和对照组,于妊娠第15天按0.3 mg/kg剂量脂多糖(lipopolysaccharide,LPS)对实验组孕鼠进行腹腔内注射,对照组孕鼠在同一天使用等量蒸馏水腹腔内注射,妊娠第21天,孕鼠均分娩,取两组新生鼠脑组织进行形态学观察和脑组织含水量测定。结果:①光镜下可见,实验组脑室周围白质水肿软化,带状或灶状凝固性坏死,而对照组无明显形态学变化;②电镜下发现,实验组少突胶质细胞线粒体肿胀明显、线粒体脊减少或空泡化,内质网明显扩张,对照组无明显变化;③实验组脑组织含水量较对照组脑组织含水量明显增高,差异具有统计学意义(P<0.05)。结论:脂多糖可导致新生鼠脑室周围白质软化,且脑组织含水量也明显增高。脂多糖是制备新生鼠脑白质软化的一种有效方法。
Objective: To confirm the reliability of lipopolysaccharide-induced neutrophil leukomalacia model. Methods: Twenty-six healthy pregnant rats were randomly divided into experimental group and control group. The pregnant rats were injected intraperitoneally with lipopolysaccharide (LPS) 0.3 mg / kg on the 15th day of pregnancy, Angelica was injected intraperitoneally with the same amount of distilled water. On the 21st day of pregnancy, all the pregnant mice were delivered. Morphological observation and brain water content were taken in the brain tissue of two neonatal rats. Results: (1) Under light microscope, the periventricular white matter edema, banding or focal coagulation necrosis were observed in the experimental group, but there was no obvious morphological changes in the control group. (2) The mitochondria swelling in the experimental group was obvious under the electron microscope, Mitochondrial ridges decreased or vacuolization, and the endoplasmic reticulum was obviously expanded. There was no significant change in the control group. (3) The water content of brain tissue in the experimental group was significantly higher than that in the control group (P <0.05). Conclusion: Lipopolysaccharide can cause neovascularization of periventricular white matter in neonatal rats, and the water content of brain tissue is also significantly increased. Lipopolysaccharide is an effective method to prepare white matter of newborn rats.
