目的 研究人脑胶质瘤组织中尿激酶型纤溶酶原激活剂 (uPA)及其受体 (uPAR)的表达 ,探讨uPA、uPAR的表达与人脑胶质瘤恶性程度的关系。方法 采用半定量逆转录—聚合酶链反应 (RT PCR)方法 ,对 49例人脑胶质瘤手术切除标本、U2 5 1等 3株胶质瘤细胞 ,12例内减压术中切除的正常脑组织标本的uPAmRNA和uPARmRNA表达水平进行检测。结果 随着胶质瘤恶性度的升高 ,其uPA及uPARmRNA表达率和表达水平逐渐增高。U2 5 1等 3株胶质瘤细胞也表达了较高水平的uPA及uPARmRNA ,而正常组织表达率及表达水平极低。结论 人脑胶质瘤中纤溶酶原激活系统活性较高 ,uPA、uPAR基因的高表达反映了胶质瘤的恶性生物学行为。
Objective To investigate the expression of urokinase-type plasminogen activator (uPA) and its receptor (uPAR) in human glioma tissue and to investigate the relationship between the expression of uPA and uPAR and the malignancy of human glioma. Methods Semi-quantitative reverse transcription-polymerase chain reaction (RT PCR) method was applied to 49 human glioma surgical specimens, U 2 5 1 and other 3 glioma cells, and 12 cases of normal excision during internal decompression. The expression levels of uPA mRNA and uPAR mRNA were detected in brain tissue samples. Results With the increase of malignant degree of glioma, the expression and expression of uPA and uPAR mRNA were gradually increased. U2 5 1 and other 3 glioma cells also expressed higher levels of uPA and uPAR mRNA, but the normal tissue expression and expression levels were extremely low. Conclusion The activity of plasminogen activation system in human gliomas is high, and the high expression of uPA and uPAR genes reflects the malignant biological behavior of gliomas.
