目的:建立甘肃不同产地1~2年生红芪和黄芪中黄酮类成分的指纹图谱,为科学评价甘肃红芪和黄芪的内在质量提供参考,并阐明红芪和黄芪能否替代使用的依据。方法:应用HPLC法,测定甘肃l-2年生各28批红芪和28批黄芪黄酮类成分指纹图谱。色谱条件:Agilent ZROBAX-C_(18)色谱柱,流动相乙腈-0.1%甲酸水溶液,梯度洗脱,流速1.0mL-min-1,检测波长254 nm,柱温35℃。结果:甘肃各产地1-2年生28批红芪有19个共有峰,28批黄芪有14个共有峰,1年生28批红芪和28批黄芪有9个共有峰,2年生28批红芪和28批黄芪有8个共有峰。甘肃各产地1年生红芪平均相似度为0.976,2年生红芪平均相似度为0.888;甘肃各产地1年生黄芪平均相似度为0.986.2年生黄芪平均相似度为0.911.1年生红芪和黄芪相似度为0.534,2年生红芪和黄芪相似度位0.490。结论:该方法重复性好,建立的指纹图谱为甘肃红芪和黄芪的质量控制及评价提供依据、随着药材生长年限的增加,甘肃红芪相似度降低,黄芪相似度降低,红芪和黄芪相似度降低。从红芪和黄芪黄酮类成分指纹图谱相似度来看,二者1年生和2年生的相似度均很低,差异较人,不适合替代使用。
OBJECTIVE: To establish the fingerprints of flavonoids from Radix Astragali and Astragalus membranaceus from 1 to 2 years old in different habitats of Gansu province, and to provide a reference for scientifically evaluating the intrinsic quality of Radix Astragali and Astragalus membranaceus in Gansu Province and elucidate the basis for the alternative use of Radix Astragali and Radix Astragali. Methods: HPLC fingerprint was used to determine the fingerprints of 28 batches of Radix Hedysari and 28 batches of Astragalus flavonoids from 1-2 years of Gansu. Chromatographic conditions: Agilent ZROBAX-C_ (18) column, mobile phase acetonitrile-0.1% formic acid aqueous solution, gradient elution, flow rate 1.0mL-min-1, detection wavelength 254 nm, column temperature 35 ℃. Results: There were 19 common peaks for 28 batches of Radix Hedysari in 1-2 years of origin in Gansu Province, 14 common peaks of 28 batches of Radix Astragali, 9 common peaks of 28 batches of Radix Astragali and 1 batch of 28 batches of Radix Astragali and 2 batches of 28 batches of Radix Astragali And 28 batches of Astragalus have 8 common peaks. The average similarity of one-year-old Radix Hedysari was 0. 976 in each area of Gansu, and the average similarity of two-year-old Radix Astragali was 0.888. The average annual similarities of one-year-old Radix Astragali were 0. 986 in Gansu Province. The average annual similarity of 2-year-old Radix Astragali was 0. 911. The similarity was 0. 534, 2-year-old Hedysari and Astragalus similarity 0.490. Conclusion: This method has good repeatability. The established fingerprint provides the basis for the quality control and evaluation of Radix Astragali and Radix Astragali. With the increase of the growth period of Radix Ginseng, the similarity of Radix Hedysari decreases and the similarity of Radix Astragali decreases. The similarity decreases. From the similarity of the fingerprint of flavonoids of Radix Astragali and Astragalus, the similarity between the two-year-old and the two-year-old is very low, the difference is rather different, which is not suitable for alternative use.
