目的研究急性肺损伤(acute lung in jury,ALI)大鼠小肠Gq/11蛋白的表达变化。方法40只健康雄性W istar大鼠尾静脉注射油酸(ole ic ac id,OA)复制ALI模型,并将其分为对照组(C组)和油酸组(OA组),再根据观测时限再将OA组分为30、60、90、120 m in 4个组。紫外法测定血管紧张肽转化酶(ACE)活性,免疫印迹法检测各组大鼠小肠黏膜的Gq/11蛋白含量。结果OA各组随时间延长Gq/11蛋白含量较C组分别增加至(138.91±23.03)%、(143.99±23.01)%、(150.68±26.25)%、(173.05±21.51)%(P<0.05或0.01)。OA组血浆和小肠LDH活性及MDA含量增高(P<0.05或0.01),血浆ACE活性较对照组降低(P<0.01),而小肠ACE活性则随时间延长逐渐增高(P<0.01)。结论Gq/11蛋白表达上调在ALI并发小肠损伤中有一定作用。
Objective To study the expression of Gq / 11 protein in small intestine of rats with acute lung injury (ALI). Methods Forty healthy Wistar rats were injected with oleic acid (OA) into the caudal vein and divided into control group (C group) and oleic acid group (OA group) The OA group was divided into 4 groups of 30, 60, 90 and 120 mins. The activity of angiotensin converting enzyme (ACE) was measured by UV method. The Gq / 11 protein content of small intestine mucosa in each group was detected by immunoblotting. Results Compared with group C, Gq / 11 protein content in the OA groups increased to (138.91 ± 23.03)%, (143.99 ± 23.01)%, (150.68 ± 26.25)%, (173.05 ± 21.51)% 0.01). Compared with the control group, the activities of plasma and intestinal LDH and the content of MDA in OA and OA in the OA group increased (P <0.05 or 0.01, P <0.01), while the activity of ACE in the small intestine gradually increased with time (P <0.01). Conclusion The up-regulation of Gq / 11 protein may play a role in ALI complicated intestinal injury.
