【目的】分析食管癌5号染色体长臂上5q23.1-q23.2区域的杂合性丢失,为食管癌候选抑癌基因定位缩小范围。【方法】用PCR银染技术,检测50例配对食管鳞状细胞癌标本6个微卫星标记的杂合性丢失情况。【结果】在检测的微卫星标记中,D5S471和D5S592杂合性丢失率高达47%和41%。【结论】D5S471和D5S592附近可能存在食管癌的候选抑癌基因。
【Objective】 To analyze the loss of heterozygosity in the region of 5q23.1-q23.2 on the long arm of esophageal cancer chromosome 5, and to narrow the range of tumor suppressor gene localization in esophageal cancer. 【Methods】 PCR-silver staining was used to detect the loss of heterozygosity of six microsatellite markers in 50 matched esophageal squamous cell carcinoma specimens. 【Result】 Among the detected microsatellite markers, the heterozygosity loss rates of D5S471 and D5S592 were as high as 47% and 41% respectively. 【Conclusion】 Candidate tumor suppressor genes of esophageal cancer may exist near D5S471 and D5S592.
