目的:观察决明子水提物对氯氮平所致大鼠体质量增加、糖脂代谢紊乱及肝脏脂质沉着的影响。方法:大鼠1~4天给予氯氮平5 mg/(kg·d),第5天起加至25 mg/(kg·d),2 w后除模型组外分别予辛伐他汀1 mg/(kg·d)或决明子水提液1、4 g/(kg·d)干预,另设正常对照组仅予生理盐水,总共给药8 w;取基线、3 d及1、2、4、6、8 w等7个时间点,称取大鼠体质量(m),测空腹血糖(FBG)及餐后2 h血糖(2h PBG);于实验第8周末采血,检测血清胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)、高密度脂蛋白(HDL-C)、果糖胺(FA)及胰岛素(INS)水平,并取大鼠肝脏进行常规HE染色。结果:实验4、6、8 w末,模型组大鼠体质量较正常对照组显著升高,2h PBG及血INS、FA、TC、TG、LDL-C升高,HDL-C降低;实验6、8 w末,决明子高、低剂量组大鼠较模型组体质量、2h PBG、INS、FA、TC、TG、LDL-C显著降低、HDL-C显著升高;肝脏病理示,模型组较正常对照组肝细胞形态紊乱,脂肪性变明显,各给药组肝组织病理学改变明显改善。结论:决明子水提物能有效纠正氯氮平所致的大鼠体质量增加、糖脂代谢紊乱及肝脏脂质沉着。
Objective: To observe the effects of aqueous extract of Cassia seed on clozapine-induced weight gain, dyslipidemia and lipid peroxidation in rats. METHODS: Clozapine (5 mg / (kg · d)) was administered in rats for 1 to 4 days and to 25 mg / (kg · d) on day 5, and after 2 weeks, simvastatin 1 mg kg / d or 1.4 g / (kg · d) of Cassia seed aqueous extract. The normal control group was given saline only for 8 weeks. The baseline, 3 days and 1, 4 , 6,8 w and other 7 time points, weighed rats body mass (m), measured fasting blood glucose (FBG) and 2 h postprandial blood glucose (2h PBG); at the end of the 8th week of blood sampling, serum cholesterol (TG), LDL-C, HDL-C, FA and INS were detected by radioimmunoassay. The liver of rats were subjected to conventional HE staining . Results: At the end of 4, 6, and 8 weeks, the body weight of rats in the model group was significantly higher than that of the normal control group. The levels of INS, FA, TC, TG, LDL-C and HDL- At the end of 8th week, the contents of PBG, INS, FA, TC, TG, LDL-C of HDL-C in 2mg high-dose and low-dose groups were significantly lower than those in model group Normal control group, morphological changes of liver cells, fatty lesions were significantly, the pathological changes of each administration group significantly improved. Conclusion: Cassia seed extract can effectively correct clozapine-induced increase in body weight, dyslipidemia and liver lipid deposition.
