观察新鲜分离和经体外培养36小时的人外周血树突状细胞DC。和DC36的形态学及其对淋巴因子与植物血凝素激活的杀伤细胞(LPAK细胞)体外诱导人肝癌细胞株BEL┐7402的促进作用。形成学观察用免疫细胞化学法。抗肿瘤实验分为三大组:(1)L组(BEL┐7402+LPAK),(2)LD36组(BEL┐7402+LPAK+DC36),(3)LD0组(BEL┐7402+LPAK+DC0)。各组均采用LPAK与BEL┐7402为5∶1和10∶1两种效靶比。培养48小时后用中性红摄入比色法检测LPAK细胞的杀伤活性。结果显示,DC36的形态以具有细长突起者为多,与DC0主要为园形或具有短而钝突起的形态有明显差别。各组的细胞毒活性依次为:LD36组>LD0组>L组(P<0.01),并随效靶比增高而相应增强。这表明新鲜分离的人外周血DC在体外经36h培养后,不仅在形态上成熟,而且抗肿瘤活性也明显增强
Freshly isolated and cultured DCs of human peripheral blood dendritic cells were observed for 36 hours. And morphology of DC36 and their promotion of human hepatocellular carcinoma cell line BEL┐7402 in vitro on lymphokines and phytohemagglutinin-activated killer cells (LPAK cells). Forming observation using immunocytochemistry. Antitumor divided into three experimental groups: (1) L group (BEL┐7402 + LPAK), (2) LD36 group (BEL┐7402 + LPAK + DC36), (3) LD0 group (BEL┐7402 + LPAK + DC0). LPAK and BEL┐7402 were used in both groups at 5: 1 and 10: 1 effect ratios. After 48 hours of culture, the killing activity of LPAK cells was examined by neutral red ingestion colorimetry. The results showed that the morphology of DC36 was more with elongated protuberances, which was obviously different from the morphology of DC0 mainly with circular or blunt protuberances. The cytotoxic activities in each group were as follows: LD36 group> LD0 group> L group (P <0.01), and increased with the increase of the target ratio. This shows that freshly isolated human peripheral blood DC after 36h culture in vitro, not only in the morphological maturity, but also significantly increased anti-tumor activity
