为了研究两种功能性海洋寡糖对S180荷瘤小鼠抗肿瘤及免疫功能的影响。建立S180荷瘤小鼠动物模型,随机分为阴性对照组(生理盐水)、阳性对照组(环磷酰胺)、寡糖不同剂量组共8组,连续灌胃10d后处死小鼠,分别计算抑瘤率、胸腺指数和脾脏指数,采用双抗体夹心ELISA法检测血清中细胞因子白介素2(IL-2)、白介素6(IL-6)、肿瘤坏死因子α(TNF-α)蛋白表达水平。结果:与生理盐水组比较,κ-卡拉胶寡糖组和壳寡糖组均能明显抑制肿瘤生长,同时两种海洋寡糖组均能够显著提高胸腺指数、脾指数、IL-2、IL-6、TNF-α的表达。提示,κ-卡拉胶寡糖与壳寡糖作用结果相似,不仅可以抑制肿瘤的生长,还可以提高荷瘤小鼠的免疫能力,其抗肿瘤作用可能与其免疫调节作用相关。
In order to study the effect of two kinds of functional marine oligosaccharides on antitumor and immune function in S180 tumor-bearing mice. The animal model of S180 tumor-bearing mice was established and randomly divided into negative control group (normal saline), positive control group (cyclophosphamide) and oligosaccharides in different dosage groups for 8 days. The mice were sacrificed after continuous gavage for 10 days, Tumor rate, thymus index and spleen index. Serum interleukin 2, interleukin 6 and tumor necrosis factor alpha (TNF-α) protein levels were detected by double antibody sandwich ELISA. Results: Compared with saline group, both kappa-carrageenan oligosaccharide and chitosan oligosaccharide groups could significantly inhibit tumor growth, meanwhile, both of the two groups could significantly increase thymus index, spleen index, IL-2 and IL- 6, TNF-α expression. These results suggest that κ-carrageenan oligosaccharides have similar effects with chitooligosaccharides, which not only inhibit the growth of tumor but also enhance the immunity of tumor-bearing mice. The antitumor effect of κ-carrageenan oligosaccharides may be related to its immunomodulatory effects.
