根据主流酶法脱墨机制,经富集、初筛和多轮复筛,成功地从造纸厂采集的土样中,分离得到了5株具有良好脱墨能力的菌株。采用液体发酵的方法,从这5株微生物中筛选得到了1株脱墨能力最好的菌株M03-3#-001。形态学观察以及ITS序列分子鉴定结果表明,该菌属于烟曲霉属。M03-3#-001菌株在100 L发酵罐上的研究结果显示,发酵时间为84h,脱墨效果最佳,白度(ISO)增加值可达到3.42,此时发酵液CMC酶活力为855.23 U/mL,α-淀粉酶活力为5 683.37 U/mL;对该菌株的脱墨条件进行研究,结果显示,脱墨反应的适宜时间为40min,适宜pH为8.0,适宜温度为30℃。
According to the mainstream enzymatic deinking mechanism, five strains with good deinking ability were successfully isolated from the soil samples collected by paper mill through enrichment, preliminary screening and multiple rounds of screening. A liquid strain M03-3 # -001 with the best deinking ability was screened from these 5 microorganisms by liquid fermentation. Morphological observation and ITS sequence molecular identification results show that the bacteria belong to Aspergillus fumigatus. M03-3 # -001 strains in 100 L fermentor on the results showed that the fermentation time was 84h, the best deinking, whiteness (ISO) value can reach 3.42, CMC enzyme activity at this time was 855.23 U / mL, and the activity of α-amylase was 5 683.37 U / mL. The deinking conditions of this strain were studied. The optimum deinking reaction time was 40 min, the optimum pH was 8.0 and the optimum temperature was 30 ℃.
