目的:观察天麻最细粉和普通粉对帕金森病(PD)大鼠神经行为学、氧化应激反应和神经炎症反应的影响,对比最细粉和普通粉的作用差别。方法:采用脑内定位注射6-羟基多巴胺(6-OHDA)建立PD大鼠模型。设立正常组,模型组,天麻普通粉低、中、高剂量组(0.45,0.9,1.8 g·kg-1)和天麻最细粉低、中、高剂量组(0.45,0.9,1.8 g·kg-1)。观察PD大鼠神经行为学、氧化应激反应和神经炎症反应的变化。结果:经过天麻普通粉和最细粉的治疗后,PD大鼠均有不同程度的神经行为学改善,丙二醛(MDA)含量降低,谷胱甘肽(GSH)含量升高,谷胱甘肽过氧化物酶(GSH-Px)和超氧化物歧化酶(SOD)活性增加,一氧化氮(NO)和α-肿瘤坏死因子(TNF-α)含量亦降低,且最细粉的效果更佳。结论:天麻普通粉和最细粉均对PD大鼠的神经行为学、氧化应激反应和神经炎症反应有改善作用,而且天麻最细粉的作用更加明显,为天麻最细粉的临床使用提供了科学依据。
OBJECTIVE: To observe the effects of the finest powder and ordinary powder of Tianma on neurobehavioral, oxidative stress and neuroinflammation in Parkinson’s disease (PD) rats, and to compare the effect of the finest powder and ordinary powder. Methods: PD rat model was established by intracerebral injection of 6-hydroxydopamine (6-OHDA). The normal group, model group, low, medium and high dose of Tianma ordinary powder group (0.45,0.9,1.8 g · kg-1) and the lowest powder of Gastrodia elata Blume low, medium and high dose group (0.45,0.9,1.8 g · kg -1). The changes of neurobehavioral, oxidative stress and neuroinflammation in PD rats were observed. Results: After the treatment of ordinary powder and the finest powder of Tianma, the PD rats all had different degree of neurobehavioral improvement, the content of malondialdehyde (MDA) decreased, the content of glutathione (GSH) increased, the content of glutathione Peptides peroxidase (GSH-Px) and superoxide dismutase (SOD) activity increased, nitric oxide (NO) and α-tumor necrosis factor (TNF-α) good. Conclusion: The ordinary powder and the finest powder of Tianma can both improve the neurobehavioral, oxidative stress and neuroinflammation of PD rats. Moreover, the effect of the fine powder of Gastrodia elata is more obvious, which provides the clinical application of the fine powder of Gastrodia elata Blume The scientific basis.
