Wortmannin是PI 3激酶的特异性抑制剂 ,它可拮抗IL 6对M 1小鼠急性髓系白血病细胞生长的抑制作用 ,但wortmannin本身对M 1细胞的生长无明显影响。凝胶阻滞电泳分析 (EMSA)表明wortmannin选择性减低IL 6对Stat3的激活作用 ,而Stat3在IL 6诱导的M 1细胞生长停止与终末分化中占重要地位。这些结果说明PI 3激酶确实参与IL 6的信号转导并参与Stat3的激活。
Wortmannin is a specific inhibitor of PI 3 kinase that antagonizes the inhibitory effect of IL 6 on M 1 mouse acute myeloid leukemia cell growth but wortmannin itself has no significant effect on M 1 cell growth. EMSA showed that wortmannin selectively reduced the activation of Stat3 by IL 6, whereas Stat3 played an important role in the termination of IL-6-induced M 1 cell growth arrest and terminal differentiation. These results indicate that PI 3 kinase is indeed involved in the signaling of IL 6 and is involved in the activation of Stat3.
