目的:采用高效液相色谱法测定大黄中大黄素、大黄酚的含量。方法:HPLC法条件:色谱柱为迪马C18(250 mm×4.6 mm,5μm),流动相为甲醇-0.1%磷酸溶液(85∶15),检测波长254 nm,流速:1.0 mL/min。结果:大黄素、大黄酚在0.009~0.72μg范围内线性关系良好(r=1),平均回收率为98.4%,RSD为0.92%。结论:方法降低了仪器要求,简化操作,准确可靠,可用于大黄清胃浓缩丸的质量控制。
Objective: To determine the content of emodin and chrysophanol in rhubarb by high performance liquid chromatography. Methods: The HPLC method was as follows: Dima C18 (250 mm × 4.6 mm, 5 μm) was used as the mobile phase. The mobile phase was methanol-0.1% phosphoric acid solution (85:15). The detection wavelength was 254 nm and the flow rate was 1.0 mL / min. Results: The emodin and chrysophanol showed good linearity (r = 1) in the range of 0.009-0.72 μg with an average recovery of 98.4% and a RSD of 0.92%. Conclusion: The method reduces the instrument requirements, simplifies the operation, is accurate and reliable, and can be used for the quality control of rhubarb and Qingwei concentrated pills.
