目的探讨消症方对子宫腺肌病在位及异位内膜间质细胞(endometrial stromal cells,ESC)增殖和凋亡的影响。方法实验分为5组:消症方高、中、低剂量组,空白对照及孕三烯酮组。采用胶原酶消化法体外培养腺肌病在位及异位ESC,药物处理腺肌病细胞24h后,采用MTT法检测药物对细胞的增殖抑制作用,显微镜观察凋亡细胞形态,采用流式细胞术检测细胞凋亡及周期。结果消症方呈一定浓度依赖性的抑制腺肌病在位及异位ESC的增殖;显微镜观察凋亡细胞形态:部分细胞固缩,胞质致密,核染色质浓集;与对照组相比,消症方能显著诱导在位及异位ESC凋亡(P<0.05);消症方可阻滞细胞周期于S期。结论消症方治疗子宫腺肌病的作用可能与其通过促进细胞凋亡、阻断周期进程以抑制腺肌病在位及异位ESC增殖有关。
Objective To investigate the effect of Xiao-Fang on the proliferation and apoptosis of eutopic and ectopic endometrial stromal cells (AEs) in patients with adenomyosis. Methods The experiment was divided into 5 groups: high, medium and low dose group, blank control and gestrinone group. The collagenase digestion method was used to culture adenomyosis in situ and ectopic ESC, and the drug-treated adenomyosis cells were treated with MTT for 24 h. MTT assay was used to detect the inhibitory effect of the drug on cell proliferation. Morphology of apoptotic cells was observed under a microscope. Flow cytometry Detection of apoptosis and cycle. Results Xiaozhengfang inhibited the proliferation of eutopic and ectopic ESCs in a concentration-dependent manner. The morphological changes of apoptotic cells were observed under a microscope: some cells were condensed, the cytoplasm was dense and nuclear chromatin was concentrated. Compared with the control group , Xiao-Fang can significantly induce eutopic and ectopic ESC apoptosis (P <0.05); Xiao-Fang can block the cell cycle in the S phase. Conclusion The therapeutic effect of Xiaozheng Decoction on adenomyosis may be related to the inhibition of adenomyosis’s eutopic and ectopic ESC proliferation by promoting apoptosis and blocking the cycle progression.
