iTRAQ技术即同位素相对标记与绝对定量技术,主要应用于蛋白质的鉴定与定量分析,结合串联质谱分析,已成为目前蛋白质组学研究的主要手段之一。因寄生虫在不同发育阶段表达虫体蛋白的差异和独特的生活史,其对宿主的选择性、致病性具有较大差异,将iTRAQ技术运用于寄生虫蛋白质组学研究,能有效找出不同阶段寄生虫表达的差异蛋白及感染后宿主表达的差异蛋白,并能对其进行高精度定量分析,这对进一步探索寄生虫生活史、研究与宿主的互作机制提供了技术支撑,同时对寄生虫病的快速诊断及治疗,寻找新的药物靶点等具有重要意义。文章简述了iTRAQ定量蛋白质组学技术及其在寄生虫病研究方面的应用进展。
The iTRAQ technology, isotope relative labeling and absolute quantification, is mainly used in the identification and quantitative analysis of proteins. Because parasites express the differences and unique life histories of the parasites at different developmental stages, they have great differences in the selectivity and pathogenicity of the host. Using the iTRAQ technology in the study of the parasite proteomics can effectively find out The differential proteins expressed by different stages of parasites and the differentially expressed proteins expressed by the host after infection can be quantitatively analyzed with high accuracy. This provides a technical support for further exploration of the life history of parasites and the interaction mechanism with the host, Parasitic diseases, rapid diagnosis and treatment, to find new drug targets and other important. The article briefly describes iTRAQ quantitative proteomics technology and its application in the research of parasitic diseases.
