目的分析苏素特异蛋白酶1(SENP-1)在脐带血造血干细胞(UCB-HSCs)中的表达情况。方法通过逆转录多聚酶连反应(RT-PCR)法检测SENP-1分子mRNA水平,以?-action为内对照;实验分为对照组A组、B组,实验组C组、D组、E组,每组检测15例标本。结果对照组中A组和B组均未表达SENP-1mRNA,实验组中C组、D组、E组均表达SENP-1类分子mRNA,以SENP-1mRNA积分光密度值/?-actin mRNA积分光密度值比较各组冻存前后及冻存时间长短SENP-1表达差异,C组为0.146±0.011,D组为0.314±0.011,E组为0.368±0.019。结论白血病细胞(HL-60)不表达SENP-1mRNA分子,即使经炎症因子r-干扰素(IFN-r)刺激也不表达,不能有效激活CD8+杀伤T细胞(CTL),是发生免疫逃逸的重要原因;脐带血造血干细胞中均有SENP-1的表达,随着冻存时间越久,SENP-1表达量越下降,表明冻存可以使脐带血干细胞的免疫原性下降。
Objective To analyze the expression of suprotein 1 (SENP-1) in cord blood hematopoietic stem cells (UCB-HSCs). Methods The level of SENP-1 mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR). The results were divided into group A, group B, group C, group D, group E , Each group detected 15 cases of specimens. Results SENP-1 mRNA was not expressed in control group A and group B, SENP-1 mRNA was expressed in group C, group D and group E, and the SENP-1 mRNA integral /? - actin mRNA The optical density values of SENP-1 were compared before and after cryopreservation and cryopreservation. The difference of SENP-1 expression in group C was 0.146 ± 0.011, in group D was 0.314 ± 0.011 and in group E was 0.368 ± 0.019. CONCLUSIONS: Leukemia cells (HL-60) do not express SENP-1 mRNA and do not express CD8 + cytotoxic T lymphocytes (CTLs) even when stimulated by IFN-γ The reason is that the expression of SENP-1 in cord blood hematopoietic stem cells was both decreased. The longer the cryopreservation period, the lower the expression of SENP-1, indicating that the immunogenicity of cord blood stem cells decreased after cryopreservation.
