目的通过对9份益母草种质资源的AFLP分析,探求各种质间的遗传多样性。方法采用扩增片段长度多态性(AFLP)标记,在DNA水平上进行遗传多样性研究,筛选了32对选择性扩增引物,将扩增出的条带作为原始矩阵,用NTSYS-PC软件计算并分析了益母草种质间的相似度,构建了遗传系统进化树。结果SDS法提取的益母草基因组DNA质量较佳,能够满足AFLP分析的要求;从32对选择性扩增引物中,筛选出10对多态性较强、带型较好、分辨率较高的组合;构建了益母草AFLP指纹图谱,将9个益母草种质全部区分开来;通过构建进化树,把9个种质分成2类。结论益母草种质资源有丰富的遗传多样性,某些形态特征差异和基因型存在相关性。
OBJECTIVE: To investigate the genetic diversity of germplasm resources in nine germplasm resources by AFLP analysis. Methods Amplified fragment length polymorphism (AFLP) markers were used to study the genetic diversity at the DNA level. Thirty-two pairs of selective amplification primers were selected and the amplified bands were used as the original matrix. NTSYS-PC software The similarity between the motherwort germplasm was calculated and analyzed, and the phylogenetic tree was constructed. Results The quality of genomic DNA extracted from motherwort by SDS was better than that of AFLP, and 10 pairs of selective amplification primers were screened for 10 pairs of genotypes with strong polymorphism, better banding pattern and higher resolution The AFLP fingerprinting of Leonurus was constructed to distinguish all 9 Leonurus species. All the 9 germplasms were divided into 2 groups by constructing phylogenetic tree. Conclusion The genetic resources of Motherwort are rich in genetic diversity, and some morphological differences are related to genotypes.
