目的:建立同时测定大鼠静脉注射荭草提取物后组织器官中荭草素、牡荆素、木犀草苷和槲皮苷含量的UPLC-MS/MS方法,研究荭草提取物在大鼠体内心、肝、脾、肺、肾等9个组织器官的分布特征。方法:色谱柱为BEH C18柱(2.1 mm×50mm,1.7μm),流动相:0.1%甲酸乙腈-0.1%甲酸水,梯度洗脱,流速0.35 mL.min-1。组织样品机械匀浆后甲醇沉淀供测定。结果:在线性范围内4个成分的线性关系良好,准确度、日内和日间精密度、提取回收率均符合生物样品的分析要求。大鼠静脉注射荭草提取物后的组织分布试验结果表明:4个成分在肾脏等9种组织器官中均能检测到。4个成分主要分布在肾、肺等组织中,在脑中分布最少。结论:本文方法可用于荭草花提取物在大鼠体内的组织分布研究。
OBJECTIVE: To establish a UPLC-MS / MS method for the simultaneous determination of tenardin, vitexin, luteolin and quercetin in the tissues and organs of rats after intravenous injection of extract of Eucheuma extract. Heart, liver, spleen, lung, kidney and other 9 tissues and organs of the distribution characteristics. METHODS: The column was BEH C18 (2.1 mm × 50 mm, 1.7 μm). The mobile phase consisted of 0.1% formic acid and 0.1% formic acid in water with a gradient of 0.35 mL · min-1. Tissue samples were mechanically homogenized after methanol precipitation for determination. Results: The four components in the linear range had good linearity, accuracy, intra-day and inter-day precision, and the extraction recovery rates were in line with the biological sample analysis requirements. Tissue distribution test results of intravenous injection of the extract of Ephedra intermedia in rats showed that four components could be detected in nine tissues and organs such as kidney. The four components are mainly distributed in the kidney, lung and other tissues, with the least distribution in the brain. Conclusion: This method can be used to study the tissue distribution of the extract of Chieh-hua flower in rats.
