目的探讨橡胶防老剂对作业人员的健康影响及可能的分子机制。方法对372橡胶防老剂生产车间作业人员及720名非接触人群进行职业健康检查,用二甲基塞唑基四唑(MTT)法检测细胞生长抑制率,用Hoechst染色法检测细胞凋亡,用免疫印迹法检测Caspase-8、Caspase-3蛋白表达。结果橡胶防老剂生产车间工人肺纹理增粗明显高于非接触组;橡胶抗氧化剂N-(1,3-二甲基)丁基-N′-苯基对苯二胺(4020)处理人胚肺成纤维细胞(HELF)24 h均可诱导细胞发生凋亡。MTT结果显示,3、6、9、12 mg/ml 4020处理HELF 24 h后,HELF细胞生长抑制率分别为8.4%、34.02%、50.37%、92.55%。3、6、9、12、15、18 mg/ml 4020引起的细胞凋亡率分别达到0.04%、1.2%、20.4%、30.8%、50.7%、90.8%;4020诱导HELF细胞凋亡过程中伴随着Caspase-3及Caspase-8蛋白表达蛋白表达增加。结论橡胶防老剂可致接触工人肺纹理增粗,体外试验提示其发生的可能机制与4020激活凋亡蛋白Caspase-3和Caspase-8有关。
Objective To investigate the health effects of rubber antioxidant on workers and possible molecular mechanisms. Methods Occupational health examination was conducted in 372 rubber antioxidant workshop and 720 non-contact groups. Cell growth inhibition rate was detected by MTT assay. Cell apoptosis was detected by Hoechst staining. Western blotting was used to detect the expression of Caspase-8 and Caspase-3 protein. Results The lung texture thickening of rubber antioxidant workshop workers was significantly higher than that of the non-contact group. The rubber antioxidant N (1,3-dimethyl) butyl-N’-phenyl-p-phenylenediamine Lung fibroblasts (HELF) can induce cell apoptosis 24 h. The results of MTT showed that the growth inhibition rates of HELF cells treated with 3, 6, 9, 12 mg / ml 4020 for 24 h were 8.4%, 34.02%, 50.37% and 92.55%, respectively. 3, 6, 9, 12, 15, 18 mg / ml 4020 could induce the apoptotic rate of HELF cells by 0.04%, 1.2%, 20.4%, 30.8%, 50.7% and 90.8% Caspase-3 and Caspase-8 protein expression increased protein expression. CONCLUSIONS: Rubber anti-oxidant can cause lung tissue thickening in exposed workers. In vitro experiments suggest that the possible mechanisms of 4020 activation are Caspase-3 and Caspase-8.
