目的:制备苦豆子总碱缓释磁性颗粒,并研究其性能。方法:采用L9(34)正交试验确定苦豆子总碱缓释磁性颗粒最佳制备工艺。以pH6.8的磷酸盐缓冲溶液为释放介质,考察其体外释放性能,并通过振动样品磁强计测定其磁性能。结果:苦豆子总碱缓释磁性颗粒最佳制备工艺组成为A1B3C2D2,即苦豆子总碱质量为1.0g,壳聚糖浓度为2.5%,壳聚糖质量为3.0g,微晶纤维素质量为1.5 g,Fe3O4纳米粒质量为1.5 g。苦豆子总碱缓释磁性颗粒具有较好的缓释性能,其体外释药特征符合一级动力学方程,同时也具有明显的磁响应性。结论:苦豆子总碱缓释磁性颗粒处方合理、制备工艺可行,具有缓释特性和超顺磁性,适用于消化道靶向给药。
OBJECTIVE: To prepare sustained release magnetic particles of total alkaloids of Sophora alopecuroides and study its properties. Methods: The optimal preparation process of total alkaloids sustained release magnetic particles of Sophora alopecuroids was determined by L9 (34) orthogonal test. The phosphate buffer solution of pH6.8 was used as the releasing medium to investigate its in vitro release performance and its magnetic properties were measured by vibrating sample magnetometer. Results: The optimal preparation process of Alkaloids from Sophora alopecuroides was A1B3C2D2. The total alkaloids content of Sophora alopecuroides was 1.0 g, the concentration of chitosan was 2.5%, the mass of chitosan was 3.0 g and the mass of microcrystalline cellulose was 1.5 g, Fe3O4 nanoparticles mass of 1.5 g. Sophora alopecuroides slow-release magnetic particles have better sustained-release properties, in vitro release characteristics in line with the first-order kinetic equation, but also has significant magnetic responsiveness. CONCLUSION: Alkaloids of Sophora alopecuroides are suitable for the targeted administration of digestive tract because of their reasonable prescription, preparation process, slow release properties and superparamagnetic properties.
