目的检测膜联蛋白A7(ANXA7)在胃癌组织中的表达改变,探讨胃癌的发生机制。方法收集人胃癌组织55例及正常胃组织25例,细胞裂解液裂解组织细胞,蛋白定量试剂盒进行蛋白定量后,Western blotting法和Real-time PCR法分别于蛋白水平和基因转录水平检测ANXA7在胃癌组织和正常胃组织中的表达改变,并用免疫组织化学法对ANXA7在细胞中的定位和表达进行检测。结果 ANXA7在胃癌中蛋白水平和转录水平的表达均显著增高,与正常组相比差异有显著性(P<0.05);ANXA7在胃癌细胞中定位于细胞质。结论膜联蛋白A7在胃癌中的表达增高有可能是胃癌发生的机制之一。
Objective To detect the expression of annexin A7 (ANXA7) in gastric carcinoma and to explore the mechanism of gastric carcinogenesis. Methods Fifty-five human gastric cancer tissues and 25 normal gastric tissues were collected. After lysed cells and protein quantification kit were used for protein quantification, Western blotting and Real-time PCR were used to detect the expression of ANXA7 at protein level and gene transcription level The expression of ANXA7 in gastric cancer tissues and normal gastric tissues was detected by immunohistochemistry. Results The expression of ANXA7 protein in gastric cancer was significantly higher than that in normal group (P <0.05). ANXA7 was located in the cytoplasm of gastric cancer cells. Conclusion The increased expression of annexin A7 in gastric cancer may be one of the mechanisms of gastric cancer.
