运用淋巴细胞杂交瘤技术,建立了4株吗啡单克隆抗体(McAb)杂交瘤细胞。分别命名为 Mor-2B_(11)、Mor—1G_3、Mor—3Ag、Mor— 4A_4。McAb 免疫学特性研究表明,4株杂交瘤分泌的抗体均为 IgG_1,亚类,Mor-2B 提纯 IgG相对亲和力为2.5ng.ml~(-1)亲和力常数为6.5×10~(-8)mol.2~(-1)与载体蛋白 OVA、BSA 及人体蛋白无交叉反应,在 McAb 与29种原型药物的交叉实验中发现海洛因,可待因有强烈的交叉反应,其余药物无明显交叉反应(抑制率<50%)。是一种很有应用价值的 McAb.
Four monoclonal antibody McAb hybridomas were established by using lymphocyte hybridoma technique. They were named Mor-2B_ (11), Mor-1G_3, Mor-3Ag and Mor-4A_4 respectively. The immunological characteristics of McAb showed that the antibodies secreted by the four hybridomas were all IgG1 subclasses. The relative affinity of Mor-2B purified IgG was 2.5 ng.ml -1 and the affinity constant was 6.5 × 10 -8 mol .2 ~ (-1) did not cross-react with OVA, BSA and human protein, and there was a strong cross-reaction between heroin and codeine in the crossover experiments between McAb and 29 kinds of archetypes, and the other drugs showed no obvious cross-reaction Inhibition rate <50%).
