目的通过观察酸脂清胶囊不同提取物对实验大鼠痛风性关节炎模型IL-1β和TNF-α的影响,研究酸脂清胶囊抗炎作用机制及对痛风性关节炎软骨破坏的作用,指导临床用药。方法酸脂清胶囊不同提取物(水提法,醇提法)大鼠灌胃7天后,尿酸钠关节腔注射造成大鼠急性痛风性关节炎模型,各组分别于造模后9h和12h随机抽取8只大鼠测量踝关节直径;下腔静脉取血,Elisa法检测炎症因子IL-1β和TNF-α。结果水提组、醇提组和秋水仙碱组致炎因子较模型组均有不同程度的降低,以水提组降低最为明显。结论酸脂清胶囊通过抑制IL-1β和TNF-α的水平,达到抗炎、保护关节软骨的作用,尤以水提组为优。
OBJECTIVE: To observe the effect of different kinds of extracts of SAC capsule on IL-1β and TNF-α in experimental rats with gouty arthritis, to study the anti-inflammatory mechanism of SAC capsule and its effect on the destruction of gouty arthritis cartilage Clinical medication. Methods The rat model of acute gouty arthritis was induced by intraperitoneal injection of sodium urate in rats after 7 days of intragastric administration of different extracts (water extraction and alcohol extraction). The rats in each group were randomized at 9h and 12h 8 rats were taken to measure the diameter of the ankle; blood was drawn from the inferior vena cava and the inflammatory cytokines IL-1β and TNF-α were detected by Elisa method. Results Compared with model group, the levels of inflammatory cytokines in water extract group, alcohol extract group and colchicine group decreased to different extents, and the decrease was most obvious in water extract group. CONCLUSION: SAPA can inhibit inflammation and protect articular cartilage by inhibiting the levels of IL-1β and TNF-α, especially in water-extract group.
