目的:观察新加下瘀血汤对糖尿病大鼠肾脏组织中转化生长因子β1(TGFβ1)和骨形成蛋白7(BMP7)表达的影响,以探讨新加下瘀血汤对糖尿病大鼠肾脏的保护作用及其机制。方法:SD大鼠随机分为正常对照组、糖尿病模型组和新加下瘀血汤治疗组。以四氧嘧啶复制糖尿病模型。12周后观察空腹血糖、血肌酐水平及24h尿微量蛋白,光镜(PAS染色)观察肾脏病理,RT-PCR技术检测肾脏组织中BMP7,TGFβ1及纤溶酶原激活物抑制因子1(PAI1)mRNA表达。结果:与模型组比较,新加下瘀血汤明显降低糖尿病模型大鼠血糖、肌酐水平和尿蛋白的排泄,肾脏病理显示肾损害明显减轻,BMP7表达明显增加,TGFβ1及PAI1 mRNA表达明显降低。结论:新加下瘀血汤对糖尿病肾脏有明显的保护作用,其机制可能与其降低肾脏TGFβ1表达,增加BMP7的表达进而降低PAI1表达有关。
OBJECTIVE: To observe the effect of Xinjiayuxuexue decoction on the expression of transforming growth factor β1 (TGFβ1) and bone morphogenetic protein 7 (BMP7) in the kidney of diabetic rats to explore the protective effect of Xinjiaxiayu Decoction on the kidney of diabetic rats Role and mechanism. Methods: SD rats were randomly divided into normal control group, diabetic model group and XJHXT treatment group. Diabetic model was replicated with alloxan. After 12 weeks, the fasting blood glucose, serum creatinine, 24h urine microalbumin and PAS staining were used to observe the renal pathology. The expressions of BMP7, TGFβ1 and plasminogen activator inhibitor 1 (PAI1) in renal tissues were detected by RT- mRNA expression. Results: Compared with the model group, XJJYX significantly reduced blood glucose, creatinine and urinary protein excretion in diabetic rats. Renal pathology showed that renal damage was significantly reduced, BMP7 expression was significantly increased, and TGFβ1 and PAI1 mRNA expressions were significantly decreased. CONCLUSION: XJXHX can obviously protect diabetic nephropathy, which may be related to decreasing the expression of TGFβ1, increasing the expression of BMP7 and then decreasing the expression of PAI1.
