目的 建立大鼠的C6胶质细胞瘤模型 ,对碳铂壳聚糖缓释微球瘤内化疗的疗效进行研究。方法 通过大鼠的C6胶质细胞瘤模型 ,用碳铂壳聚糖缓释微球进行瘤内化疗 ,检测药物的疗效和动力学分布。结果 随着药物从微球中的释放 ,药物的浓度逐渐上升 ;瘤心浓度最高 ,边缘次之 ,瘤旁脑组织更低 ;碳铂在肾脏及血浆内的分布是一致的 ,但较其在脑内的浓度相差甚远 (P <0 0 0 0 1) ;病理检查显示投药后肿瘤细胞的生长受到明显的抑制 ,并有明显的剂量依赖性 ,Giemsa染色肿瘤细胞出现特征性的凋亡细胞群。结论 碳铂壳聚糖缓释微球有效地克服了纯碳铂制剂的不足 ,瘤内化疗效果佳 ,全身、局部毒副反应均小 ,具有很好的临床前景。
Objective To establish a rat C6 glioma model and study the therapeutic effects of intraperitoneal chemotherapy with carbon microspheres on chitosan sustained release microspheres. Giemsa staining showed characteristic apoptotic cells in tumor cells. group. Conclusion The carbon platinum chitosan sustained-release microspheres can effectively overcome the deficiency of pure carbon platinum preparations. The intratumoral chemotherapy is effective, and the systemic and local toxicity are all small. It has a good clinical prospect.
