以从杨树发病叶分离到的病原菌为试材,采用形态学与分子生物学法鉴定病原菌,利用菌丝培养法,研究不同培养基、温度、光照、pH值、碳源和氮源对病原菌菌落生长的影响。结果表明:病原菌为大茎点属(Macrophomasp.)真菌;病原菌菌丝生长最适温度为25~30℃,适宜pH值为6.0,适宜培养基为PA培养基与PDA培养基,不同的光照条件对病原菌菌丝生长速度影响不大,适宜病原菌P1生长的最佳碳源为乳糖,氮源为酵母膏。
