目的:确定新城鸡瘟病毒Losota系(NDV-L)能否诱导入外周血单核细胞(PBMCs)产生一氧化氮(NO)以及该NO是否与 PBMCs介导的细胞毒作用有依赖关系.方法:Griess、FACS分析和~3H-TdR同位素释放法.结果:NDV-los不同时间(1,2,4,8,12,24和48小时)与人PBMCs作用后:发现NDV-L与PBMCs作用,FACS分析显示吸附率从2小时以后均在75%以上;如果收集NDV-L与PBMCs作用24小时的上清,用Griess方法则可检测到NO,说明NDV-L可以诱导PBMCs释放NO,且释放量(72μM)与阳性对照组(BCG-LPS作用的
Objective: To determine if NDV-Losota strain (NDV-L) can induce the production of nitric oxide (NO) in peripheral blood mononuclear cells (PBMCs) and whether this NO is dependent on the cytotoxicity mediated by PBMCs.Methods: Griess, FACS analysis and ~ 3H-TdR isotope release method.Results: NDV-L and PBMCs treated with different concentrations of NDV-los (1, 2, 4, 8, 12, 24 and 48 hours) FACS analysis showed that the adsorption rate was above 75% after 2 hours.
