目的 观察重组防御素 5 (r HD- 5 )对 6株肠道常驻细菌的杀菌效果。方法 通过冷冻干燥获得含有 r HD- 5的细胞培养上清 ( 液 )、5倍浓缩液 ( 液 )和 10倍浓缩液 ( 液 ) ,以不含 r HD- 5的培养上清 (O液 )为对照 ,观察了它们对大肠埃希菌 (ATCC 2 5 92 2 )、金黄色葡萄球菌 (ATCC 2 5 92 3)、铜绿假单胞菌 (ATCC 2 785 3) ,表皮葡萄球菌、铜绿假单胞菌、鲍氏不动杆菌野生株的抑菌环直径大小。结果 随着 r HD- 5浓度的逐渐增加 ,O、 、 、 液对大肠埃希菌的抑菌环直径分别为 0 mm、(15 .78± 0 .6 7) m m、(31.11± 0 .89) m m、(36 .94± 0 .77) mm ;对金黄色葡萄球菌 (ATCC 2 5 92 3)的抑菌环直径分别为 0 mm、 (18.6 7± 0 .35 ) mm、(2 7.11± 0 .5 5 ) mm、(30 .72± 0 .4 4 )mm ;仅 液对铜绿假单胞菌 (ATCC 2 785 3)产生了抑菌环 (16 .6 7± 0 .79mm ) ;O、 、 、 液对表皮葡萄球菌野生株的抑菌环直径分别为 0 m m、0 mm、(15 .6 7± 0 .5 0 ) m m、(30 .5 6± 0 .5 8) mm ;而仅有 液对鲍氏不动杆菌野生株产生了抑菌环 (14 .4 4± 0 .77) m m ,对铜绿假单胞菌野生株均没有抑菌环产生。结论 r HD- 5对 6种肠道常驻细菌具有不同程度的杀菌活性 ,提示其在防治肠源性感染中具有潜在的临床应用价?
Objective To observe the bactericidal effect of recombinant defensin 5 (r HD-5) on six strains of intestinal bacteria. Methods Cell culture supernatant containing r HD-5, 5 times concentrated solution (liquid) and 10 times concentrated solution (liquid) were obtained by lyophilization. The culture supernatant (liquid O) without r HD- As controls, their effect on Escherichia coli (ATCC 2 5 92 2), Staphylococcus aureus (ATCC 2 5 92 3), Pseudomonas aeruginosa (ATCC 2 785 3), Staphylococcus epidermidis, Bacillus, Acinetobacter baumannii inhibition of the diameter of the antibacterial ring. Results With the increasing of r HD-5 concentration, the diameters of antibacterial rings of O, V and L against Escherichia coli were 0 mm, (15.78 ± 0.67) mm and (31.11 ± 0.89 ) and (36.994 ± 0.77) mm, respectively. The diameters of bacteriostatic rings against Staphylococcus aureus (ATCC 2 5 92 3) were 0 mm, (18.6 7 ± 0.35) mm, (2 7.11 ± 0 .5 5) mm, (30.72 ± 0.44) mm; only the bacteriolytic ring (16.67 ± 0.79mm) was produced on Pseudomonas aeruginosa (ATCC 2 7853); O The antibacterial ring diameters of the wild strains of Staphylococcus epidermidis were 0 mm, 0 mm, (15.67 ± 0.500) mm and (30.5 ± 0.58) mm, respectively. However, The only solution had a bacteriostatic ring (14.4 ± 0.77) mm on the wild strain of Acinetobacter baumannii and no antibacterial ring on the wild-type of Pseudomonas aeruginosa. Conclusion r HD-5 has different degrees of bactericidal activity against six kinds of intestinal bacteria, suggesting that it has potential clinical application in the prevention and treatment of intestinal infection.
