已知膀胱内灌注αTNF(肿瘤坏死因子)对局限性膀胱癌有较好的控制作用。近年有报道加全身用γIFN(干扰素)可增加αTNF的细胞毒作用,这已在我们所作的体外试验中得到证实。本研究是观察αTNF用逐步加量或更频繁的用药方案是否可增加其作用,加用IFN是否可取得额外的效果。材料和方法: 用18~20克的雌鼠,致癌后,随机分成多组,每组25~30只。用γIFN组是在每次膀胱灌注前,静脉推注1.3ug/只/次(30,000u)。膀胱灌注αTNF分两种方案:①12.5ug/只/次,分别在致癌后第77、11、15天灌注。②2.5ug/只/次,在第7、911、13、15天灌注。制剂用PBS溶解,
It is known that intravesical instillation of αTNF (tumor necrosis factor) has a better control of localized bladder cancer. In recent years, it has been reported that addition of systemic IFN (interferon) can increase the cytotoxic effect of αTNF, which has been confirmed in our in vitro experiments. This study is to observe whether the use of gradual or more frequent administration of αTNF can increase its effect, and whether additional IFN can achieve additional effects. Materials and Methods: 18 to 20 grams of female mice were used. After carcinogenesis, they were randomly divided into groups of 25 to 30. The γIFN group was given an intravenous bolus of 1.3 ug/mouse/time (30,000 u) before each bladder infusion. Intravesical infusion of αTNF was divided into two groups: 112.5ug/rat/times, which were injected on the 77th, 11th and 15th days after carcinogenesis. 22.5ug/only/times, perfused on days 7, 911, 13 and 15. The preparation was dissolved in PBS.
